You can enable best offer in Sales Manager Pro from the "eBay Site Information" tab (when creating a listing) by checking the box for "Enable best offer for this listing" near the top right (immediately below where you can enable auto-relist). Please let us know should you continue to have trouble with this.

I see where you are supposed to be able to check the box for best offer, but mine is grayed in and can't be checked.
What type of listing are you creating? Note that the best offer option cannot be used with auction listings (it can only be used with fixed price listings). After you have selected to create a fixed price listing, you will also need to select your category before you can select the best offer option.
